چکیده

A B S TRA CT The transfer of normal human fibroblasts from medium containing whole serum to medium devoid of lipoproteins produced a 90% decrease in the cellular content of cholesteryl esters and a 30% decrease in the free cholesterol content. When these lipoproteindeprived cells were subsequently incubated with human low density lipoprotein (LDL), there was a 7-fold increase in the cellular content of esterified cholesterol and a 1.6-fold increase in the cellular content of free cholesterol. The concentration at which LDL produced its half-maximial effect in elevating cellular sterol content (30 yg/ml of LDL-cholesterol) was similar to the half-maximal concentration previously reported for high affinity binding of LDL to its cell surface receptor. High density lipoprotein (HDL) and whole serum from a patient with abetalipoproteinemia (neither of which contains a component that binds to the LDL receptor) did not produce a significant increase in the content of either cholesterol or cholesterYl esters in normal cells. Furthermore, in fibroblasts from patients with the honmozygous form of familial hypercholesterolemia, which lack functional LDL receptors, LDL had no effect in raising the cellular content of either free or esterified cholesterol even when present in the medium at concentrations as high as 450 iyg sterol/nml. It is concluded that LDL-receptor interactions constitute an important biochemical mechanism for the regulation of the cholesterol content of normal human fibroblasts. Mforeover, when considered in light of current concepts of LDL metabolism in intact mammnals, the present data suggest that a major function of

Chronic Aerobic Exercise Decreases Lectin-Like Low Density Lipoprotein (LOX-1) Receptor Expression in Heart of Diabetic Rat

Background: Overexpression of lectin-like low density lipoprotein (LOX-1) receptor plays an important role in hyperglycemia-induced vascular complications such as atherosclerosis. Based on the beneficial effects of exercise on preventing cardiovascular complications of diabetes, we aimed to examine the protective effects of aerobic exercise on expression of LOX-1 receptor and production of free...

Familial Hypercholesterolemia in Iran: A Novel Frameshift Mutation in Low Density Lipoprotein Receptor (LDLR) Gene

Background and Objective: Familial hypercholesterolemia (FH) is an autosomal trait, which is caused by mutations in Low Density Lipoprotein Receptor (LDLR) gene. FH penetrance is about 100% and worldwide prevalence for heterozygous subjects is almost 1 in 500 and for homozygous 1 in 1,000,000. The patients are at risk of premature coronary heart disease (CHD) due to defective LDLR a...

Compare the Effect of Eicosapentaenoic Acid and Oxidized Low-Density Lipoprotein on the Expression of CD36 and Peroxisome Proliferator-Activated Receptor Gamma

Background: There is evidence that CD36 promotes foam cell formation through internalizing oxidized LDL (ox-LDL) into macrophages therefore, it plays a key role in pathogenesis of atherosclerosis. In addition, CD36 expression seems to be mediated by nuclear receptor peroxisome proliferator-activated receptor gamma (PPAR-γ). The aim of the present study was to evaluate and compare the effect of ...

A Novel Mutation in Exon 4 of the Low Density Lipoprotein (LDL) Receptor Gene in an Iranian Familial Hypercholesterolemia Patient

Familial hypercholesterolemia (FH) is an autosomal co-dominant disorder of lipid metabolism, caused by mutations in LDL receptor gene. The penetrance of FH is almost 100%, meaning that half of the offspring of affected parents born with disease. The patients are at risk of premature coronary heart disease (CHD). There is no report about the molecular basis of FH in Iran. Identification of mutat...

Attenuating Effect of Curcumin on Diet-induced Hypercholesterolemia in Mice

Background and Aims: Atherosclerosis is currently a chronic disease in which cholesterols accumulate in large arteries. Many genes such as liver X receptor α (LXRα) are involved in the cholesterol homeostasis. Curcumin, the main active polyphenol component derived from Curcuma longa, contribute to anti-inflammation and antioxidant in the treatment of atherosclerosis. Thus, this stud...

In Vitro Effect of ?-Tocopherol, Ascorbic Acid and Lycopene on Low Density Lipoprotein Glycation

Nonenzymatic glycation of low density lipoprotein (LDL) is a reaction of glucose and other reducing sugars with apolipoprotein B100 (apo-B100) lysine residues. In diabetes, this reaction is greatly accelerated and is important in the pathogenesis of diabetic complications. The objective of this study was to investigate in vitro effects of ?-tocopherol, ascorbic acid and lycopene on LDL glycatio...
